NYC nanny pleads not guilty in deaths of two kids

NEW YORK (Reuters) - A New York nanny accused of the stabbing deaths of two young children pleaded not guilty to murder charges Wednesday in a judicial proceeding conducted in the hospital where the 50-year-old woman is handcuffed to her bed as she recovers from self-inflicted wounds.

Yoselyn Ortega was indicted two weeks ago on two counts each of first- and second-degree murder in the killings of Lucia Krim, 6, and her brother, Leo, who was days shy of his second birthday, in their parents' luxury apartment.

Ortega was under a white blanket, handcuffed to her hospital bed and wearing a blue hair net, according to a pool report of the proceeding at New York-Presbyterian/Weill Cornell Medical Center. She appeared alert but did not speak during the 10-minute proceeding.

Ortega's attorney, Valerie Van Leer-Greenberg, entered the plea on her behalf, the pool report said. Judge Lewis Bart Stone ordered Ortega held without bail while she undergoes a psychiatric exam. The next court date was set for January 16.

Ortega has been in the hospital since October 25, recovering from what police have described as stab wounds she inflicted on herself at the scene of the murders.

The children's mother, Marina Krim, discovered their bodies in the bathtub when she returned to their Upper West Side home from a swimming lesson with a third child, police have said. Ortega then began to stab herself in front of Krim.

"Yoselyn Ortega is charged with taking the lives of two innocent children who were incapable of defending themselves," New York District Attorney Cyrus Vance said in a statement. "This crime shocked and horrified parents around the city, many of whom entrust their children to the care of others both by necessity and by choice."

Ortega was formally arrested earlier this month after police interviewed her for the first time in her hospital room.

Ortega had been employed by the Krim family for two years, police said. She lived with her son and sister near the Krims' apartment off Central Park and has been a naturalized U.S. citizen for a decade.

Marina Krim had expected to meet Ortega and the two children at a dance studio on the day of the killings. Upon finding the lights out in the apartment, Marina Krim asked the doorman if Ortega had taken the kids out, police said.

Krim then re-entered the home and found the children in the bathroom before encountering Ortega.

Facebook Copyright Post a Hoax | XFINITY News Blog by Comcast

A new post showing up on Facebook has some users incorrectly thinking they have copyright ownership of the images and content they post to the social media site.

The New York Daily News calls the post a ?hoax,? and says that users are still beholden to Facebook?s sharing policy despite any legal notice posted.

Many of the posts start out with a section of text that reads:

?In response to the new Facebook guidelines I hereby declare that my copyright is attached to all of my personal details, illustrations, comics, paintings, professional photos and videos, etc. (as a result of the Berner Convention). For commercial use of the above my written consent is needed at all times!?

Snopes.com, a site dedicated to sniffing out and debunking online hoaxes, also calls the posting false.

?In fact, there are no ?new Facebook guidelines,? the ?Berner Convention? is a bungled reference to the Berne Convention ? a 126-year-old international copyright policy ? and users are still bound by the terms and conditions set by Facebook during sign up,? the Daily News reports.

Snopes said the posting is similar to another hoax that circulated several years ago when website owners believed they could avoid piracy prosecution by posting a legal notice on sites.

?Facebook users cannot retroactively negate any of the privacy or copyright terms they agreed to when they signed up for their Facebook accounts nor can they unilaterally alter or contradict any new privacy or copyright terms instituted by Facebook simply by posting a contrary legal notice on their Facebook walls,? Snopes reports.

NAIROBI, Kenya, Nov 26 ? Chief Justice Willy Mutunga says an International Crimes Division of the High Court could be in place as early as next month to deal with medium and low level perpetrators of grave crimes in the country.

Mutunga who spoke at a training program for Kenyan journalists on the International Criminal Court processes, said that among other things, the division will be focusing on crimes like those against humanity and others.

?The security, stability and economy of this nation is mortally threatened by crimes against humanity, drug trafficking, trafficking in humans and body parts, money laundering, counterfeit goods, corruption, piracy and terrorism,? he said.

He emphasised the need for these crimes to be dealt with expeditiously.

?Something must be done and the ICD (International Crimes Division) is the Judiciary?s response to it. The ICD project is owned by the Council for the Administration of Justice, the assembly line for the justice sector. It has robust support of the Ministry of Justice, the AG, the DPP, the Police Commissioner, the prisons, other security services, private sector, civil society and grassroots media.?

He however pointed out that the ICD has nothing to do with Kenyan cases pending before the ICC.

German Ambassador Margit Hellwig also welcomed the establishment of the International Crimes Division saying that it will bring justice to all Kenyans.

She further called for enhanced civic education to be carried out to demystify the process.

Attorney General Githu Muigai who spoke at the same venue insisted that Kenya is cooperating fully with the International Criminal Court.

?Since the case against Kenyan suspects was opened and since I have been sitting in the AG?s office we have cooperated fully with the court they have material after material after material,? he pointed out.

The AG stated that Kenya, will not listen to those who shout loudest about the ICC and are not member states of the ICC.

?The loudest commentators on the ICC and its importance to the future international justice are not members of the Rome Statute. The united States of America is not a member of the Rome Statute although it has an International Ambassador on international crimes who has been to my office to encourage me to follow the Rome statute,? he said.

Deputy Prime Minister Uhuru Kenyatta, Eldoret North MP William Ruto, former civil service chief Francis Muthaura as well as radio presenter Joshua arap Sang are due to appear before the ICC in April next year, when trials against them over the 2008 post-election violence are set to kick off.

(AP) ? Americans clicked away for deals on Cyber Monday, which is expected to be the biggest online shopping day in history.

It's estimated that this year's Cyber Monday will be the biggest online shopping day of the year for the third year in a row: According to research firm comScore, Americans are expected to spend $1.5 billion, up 20 percent from last year on Cyber Monday.

Early results show that online shopping was up 24.1 percent compared with the same time period a year ago, according to figures by IBM Benchmark released at noon. Sales from mobile devices, which include tablets, rose 12 percent. The group does not track dollar amount sales.

The strong start to Cyber Monday, a term coined in 2005 by a shopping trade group that noticed people were doing a lot of shopping on their work computers on the Monday following Thanksgiving, comes after overall online sales rose during the four-day holiday shopping weekend that began on Thanksgiving.

"Online's piece of the holiday pie is growing every day, and all the key dates are growing with it," said Forrester Research analyst Sucharita Mulpuru. "The web is becoming a more significant part of the traditional brick-and-mortar holiday shopping season."

It's the latest sign that Americans are becoming addicted to the convenience of the Web. With the growth in smartphones and tablet computers, shoppers can buy what they want, whenever they want, wherever they want. As a result, retailers have ramped up the deals they're offering on their websites during the holiday shopping season, a time when stores can make up to 40 percent of their annual revenue.

Amazon.com, which started its Cyber Monday deals at 12:01 a.m. Monday, is offering as much as 60 percent off a Panasonic VIERA 55-inch TV that's usually priced higher than $1,000. Sears is offering $430 off a Maytag washer and dryer, each on sale for $399. And Kmart is offering 75 percent off all of its diamond earrings and $60 off a 12-in-1 multigame table on sale for $89.99.

Delisa O'Brien, 24, took advantage of some of the deals on Monday. O'Brien, who said she would rather shop online than deal with the crowds in stores, bought an H-P Notebook for $399 on Hewlett Packard's website for her mother. The company threw in a free Nook e-book reader with her purchase.

"When it comes to Black Friday, I'm a tiny, 5'1" woman and the thought of having to push and shove my way through hoards of people just to get cheap merchandise is kind of a nightmare to me," said O'Brien, a Brooklyn, N.Y. resident. "My mom gets a new laptop, I get an e-reader, and all without spending too much money ... Everybody wins."

Jack Payne, 60, an emergency management worker in St. Lucie County, Fla., shopped on Cyber Monday for the first time, picking up a refurbished iPad on apple.com for $469, down from a new iPad price of $599. He compared prices on Amazon.com and Best Buy's website but found the best deal on the Apple website.

"The economy still sucks so every penny counts," he said.

How well retailers fare on Cyber Monday will offer insight into Americans' evolving shopping habits during the holiday shopping season. With the growth in high speed Internet access and the wide use of smartphones and tablets, people are relying less on their work computers to shop than they did when Shop.org, the digital division of trade group The National Retail Federation, introduced the term "Cyber Monday."

As a result, the period between Thanksgiving and Cyber Monday has become busy for online shopping as well. Indeed, online sales on Thanksgiving Day, traditionally not a popular day for online shopping, rose 32 percent over last year to $633 million, according to comScore. And online sales on Black Friday were up 26 percent from the same day last year, to $1.042 billion. It was the first time online sales on Black Friday surpassed $1 billion.

For the holiday season-to-date, comScore found that $13.7 billion has been spent online, marking a 16 percent increase over last year. The research firm predicts that online sales will surpass 10 percent of total retail spending this holiday season. The National Retail Federation estimates that overall retail sales in November and December will be up 4.1 percent this year to $586.1 billion.

New 3-D movie technology: are viewers ready?

When the?3-D version of "The Hobbit: An Unexpected Journey" debuts in movie theaters on Dec.?14, the film will likely wow viewers with its special effects. But audiences at select theaters will get even more bang for their buck: they?ll be the first to witness a new technology that displays images at a rate of 48 frames per second, twice as fast as the current standard.

The new display rate is sure to look different. Whether audiences will like it is less certain. So far, early reviews have been mixed, according to 3D Focus, a website that tracks changes in 3-D-viewing technology.?

Some people think the effects look "too real." For others, eyestrain, headaches, nausea and other discomforts are as much of a hazard as they were with the older technologies. That's because most films that are designed to make images look like they are popping out of the screen work by showing each eye different pictures at different times. That forces the brain to merge conflicting messages -- an obstacle that 3-D studios are still working to overcome.

"People really differ," said Marty Banks, a vision scientist at the University of California, Berkeley. "Some people just don't get it and they're fine. Other people are fairly uncomfortable. The industry is really worried about this. They're listening to us?scientists and they?re taking our advice."

ll movies, whether "2-D" or "3-D" create the illusion of movement by flashing one still image after another onto the screen at a rate that is fast enough to fool our brains into seeing action. If the sequence of images is presented too slowly, the result is a shaky effect called "judder" that can sometimes look like a double image.

3-D technology (which Banks prefers to call stereo-3-D or S3-D because even "2-D" movies look three-dimensional) raises the risk for judder because these films usually show screen shots in an alternating sequence to one eye at a time. First the left eye sees an image, then the right eye sees one, then the left eye sees the next image, and so on.

With the latest generation of 3-D glasses, polarizing filters allow each eye to see light coming from just one direction, and screen shots are displayed so that a single lens is targeted at a time. When one eye is stimulated, the other eye is presented with darkness.

It's up to the brain to put everything together into a coherent picture. But with each eye seeing alternating flashes of brightness and darkness, jumpy judder can creep in, causing distraction or discomfort.

To solve that problem, 3-D films that are shot at a standard 24 frames per second actually show the left eye a single image three times in a row with flashes of darkness in between. Then the right eye gets its image three times in a row, and that pattern continues for the entire film. This "triple flashing" allows the screen to display 72 frames per second, which reduces flicker effects.

For the new Hobbit, director Peter Jackson used new technology to film at 48 frames per second instead of 24. In real life, our visual systems take in information continuously, and some experts speculate that images will need to be captured at more than 100 or even 150 frames per second before a movie looks truly real. But with any increase in frame-rate speed, motion should look smoother and more realistic.

"As you increase the rate of new incoming data, you're getting closer and closer to the real world," Banks said. "At some rate, you will get to the point where you can?t tell the difference."

But first, moviemakers have some technical issues to deal with.

One question, Engle said, is whether the technological limitations of the projectors used by most theaters will require them to use fewer flashes for each eye before switching to the other eye?s image????a switch that could actually cause new stuttering problems.

Some theaters use a dual-projector set-up that gets around that issue by showing images to both eyes at the same time, and in those cases, Engle said, the results should be spectacular. But theaters that use a single projector to display 3-D movies will have some new decisions to make, and audiences might not be universally happy about the final product.

"You have to be careful what you wish for," said Rob Engle, 3-D supervisor at a major film studio in Burbank, Calif. "If you have to make compromises in order to increase the frame rate by decreasing the flash rate, you start to run into trade-offs."

As higher frame rates lead to more realism, dismay is also cropping up among people who are used to seeing films that look like films rather than television or real life. Similar complaints have emerged whenever filmmaking technology has risen to a new level over the decades.

"Every time we've moved closer to reality by adding sound, adding color or getting rid of scratches, in the end, the audience has accepted it and there's no going back," Banks said. "I think the same is going to be true here.

"In the end, we all want immersion. We want to feel like we're in that scene and closer to reality. I think once people see it, they are going to go, 'Wow. I want that.'"

Jolla shows off Sailfish mobile OS, promises 'effortless multitasking,' speed galore

As promised, Jolla has shown off its newly launched Sailfish OS today, and said that UI will be all about speed and multitasking, while also cutting down on the finger dancing required to use it. The experience will start from the homescreen, where users will be able to control music playback or end a call using a "pulley" menu system without needing to open the respective apps. The Finnish startup also said that "many" Android apps would run on Sailfish handsets without modification, though a simple port would be required for best operation. Jolla foresees the software running not only smartphones from hardware players like new partner ST-Ericsson, but also tablets, smart TVs and in-car systems, for instance. Violence flares as Clinton seeks Gaza truce

GAZA/JERUSALEM (Reuters) - U.S. Secretary of State Hillary Clinton on Wednesday pursued a Gaza truce, with Israel and Hamas still at odds over key terms, as Israeli air strikes shook the enclave and a bomb exploded on a Tel Aviv bus.

After talks in Ramallah with Palestinian President Mahmoud Abbas, Clinton held a second meeting with Israeli Prime Minister Benjamin Netanyahu before heading to Egypt, the main broker in efforts to end eight days of fighting and avert a possible Israeli ground offensive.

In Tel Aviv, at least 10 people were wounded when a bus was blown up on a main street near the Defence Ministry and military headquarters. Israel's government called it a terrorist attack.

The explosion, which police said was caused by a bomb placed on the vehicle, touched off celebratory gunfire from militants in Gaza and threatened to complicate truce efforts.

Israel's best-selling Yedioth Ahronoth newspaper said an emerging outline of a ceasefire agreement called for Egypt to announce a 72-hour ceasefire followed by further talks on long-term understandings.

Under the proposed document, which the newspaper said neither party would be required to sign, Israel would hold its fire, end attacks against top militants and promise to examine ways to ease its blockade of the enclave.

Hamas, the report said, would pledge not to strike any Israeli target and ensure other Palestinian factions in the Gaza Strip also stop their attacks.

An Israeli political source said differences holding up a deal centered on a Hamas demand to lift the Gaza blockade completely and the kind of activity that would be allowed along the frontier, where Israeli troops often fire into the enclave to keep Palestinians away from an area near a border fence.

Hamas official Ezzat al-Rishq said the main stumbling block was "the temporary timeframe for a ceasefire that the Israelis want us to agree to".

"GOOD INTENTIONS"

The London-based Al Hayat newspaper, citing sources in Hamas and Islamic Jihad, said Israel wanted a 90-day period to determine "good intentions" before discussing Palestinian demands, a position the report said the groups have rejected.

Rishq said a short-term truce, whose proposed duration he did not disclose, "would only buy (Israel) time" until a general election in January and "we would have accomplished nothing in the way of a long-term truce".

Hamas sources said the group was also demanding control over Gaza's Rafah borders with Egypt, so that Palestinians could cross easily, and Israeli guarantees to stop assassinating Hamas leaders.

Israel, one of the Hamas sources said, wanted a commitment from the group to stop smuggling through tunnels that run into Gaza under the Egyptian border. The tunnel network is a conduit for weapons and commercial goods.

News of the Tel Aviv bus bombing, the first serious blast in Israel's commercial capital since 2006, emerged just after Clinton and Netanyahu ended their meeting. A spokesman for Netanyahu declined to give details on their discussions, which followed talks on Tuesday.

Clinton, who flew to the region from an Asian summit, said in her public remarks after Tuesday's meeting with Netanyahu that it was "essential to de-escalate the situation".

"The rocket attacks from terrorist organizations inside Gaza on Israeli cities and towns must end and a broader calm restored," she said.

Clinton earlier assured Netanyahu of "rock-solid" U.S. support for Israel's security, and praised Egyptian President Mohamed Mursi's "personal leadership and Egypt's efforts thus far" to end the Gaza conflict and promote regional stability.

"As a regional leader and neighbor, Egypt has the opportunity and responsibility to continue playing a crucial and constructive role in this process. I will carry this message to Cairo tomorrow (Wednesday)," she said.

"LONG-TERM" SOLUTION

Netanyahu told Clinton he wanted a "long-term" solution. Failing that, Netanyahu made clear, that he stood ready to step up the military campaign to silence Hamas' rockets.

"A band-aid solution will only cause another round of violence," said Ofir Gendelman, a Netanyahu spokesman.

While diplomatic efforts continued, Israel struck more than 100 targets in Gaza overnight, killing a Hamas gunman and destroying a cluster of Hamas government buildings.

Palestinians militants fired 31 rockets at Israel, causing no casualties, and Israel's Iron Dome interceptor system shot down 14 of them, police said.

Israel has carried out more than 1,500 strikes since the offensive began. Medical officials in Gaza said 139 Palestinians, most of them civilians, including 34 children, have been killed. Nearly 1,400 rockets have been fired into Israel, killing four civilians and a soldier, the Israeli military said.

In the West Bank city of Ramallah, Clinton held talks with Palestinian President Abbas, whose bid to upgrade the Palestinians' status at the United Nations, in the absence of peace negotiations with Israel, is opposed by Washington.

"Secretary Clinton informed the president that the U.S. administration is exerting every possible effort to reach an immediate ceasefire and the president expressed his full support for this endeavor," said Palestinian chief negotiator Saeb Erekat.

"Once the Israelis accept to stop their bombardments, their assassinations, there will be a comprehensive ceasefire sustained from all parties," Erekat said.

A Palestinian official with knowledge of Cairo's mediation told Reuters that Egyptian intelligence officials would hold further discussions on Wednesday with leaders of Hamas and the Islamic Jihad group.

"There may be a response from Israel that Egyptian mediators want to present to Hamas and Islamic Jihad leaders," the official said. "Let's be hopeful it would be something Palestinian factions can accept."

Like most Western powers, Washington shuns Hamas as an obstacle to peace and has blamed it for the Gaza conflagration. A U.N. Security Council statement condemning the conflict was blocked on Tuesday by the United States, which complained that it "failed to address the root cause," the Palestinian rockets.

Hamas for its part is exploring the opportunities that last year's Arab Spring has given it to enjoy favor from new Islamist governments, and from Sunni Gulf powers keen to woo it away from Shi'ite Iran.

It may count on some sympathy from Mursi, although Egypt's first freely elected leader, whose Muslim Brotherhood inspired Hamas' founders, has been careful to stick by the 1979 peace deal with Israel struck by Cairo's former military rulers.

Along the Gaza border, Israeli tanks, artillery and infantry remained poised for a possible ground offensive in the densely populated enclave of 1.7 million Palestinians.

But an invasion, likely to entail heavy casualties, would be a major political risk for Netanyahu, who is currently favored to win the upcoming Israeli election. More than 1,400 Palestinians were killed in Israel's three-week war in the Gaza Strip in 2008-9, prompting international criticism of Israel.

Education for parents in Taranaki is being taken a step further thanks to support from community groups.

The New Plymouth West Rotary Club presented Plunket Taranaki with $5600 last night to put towards its parenting education programme. Plunket community services leader Leigh Morrice said a need for a parenting education programme in South Taranaki had been identified, particularly in the Stratford area.

"We have been recruiting facilitators and now have a programme in progress out there," she said.

"To implement these types of programmes we have to rely on funding, so every little bit helps and we're very appreciative of Rotary."

About 20 Rotary members were given a tour of Plunket's newly-opened premises last night, and president Michael Archer said he was more than happy to hand over the cheque.

"They're an extremely worthwhile local charity who have helped so many families over the years, and we are very happy to help them further," Mr Archer said.

The family support service relocated from its old Courtenay St building to an eco-friendly Taranaki area headquarters at 214 Devon St West in September.

Mrs Morrice said the new facility operated two clinics, had meeting rooms used for community groups and anti-natal classes, and also housed Plunket's car seat initiative.

In the New Plymouth area about 520 babies born each year received a free service from Plunket, she said.

About 90 per cent of those families used the service.

Each day the centre would see about 30 or 40 families, either at home by one of the centre's seven nurses, or at the new facility.

3-D light switch for the brain: Device may help treat Parkinson's, epilepsy; aid understanding of consciousness

ScienceDaily (Nov. 19, 2012) ? A new tool for neuroscientists delivers a thousand pinpricks of light to a chunk of gray matter smaller than a sugar cube. The new fiber-optic device, created by biologists and engineers at the Massachusetts Institute of Technology (MIT) in Cambridge, is the first tool that can deliver precise points of light to a 3-D section of living brain tissue. The work is a step forward for a relatively new but promising technique that uses gene therapy to turn individual brain cells on and off with light.

Scientists can use the new 3-D "light switch" to better understand how the brain works. It might also be used one day to create neural prostheses that could treat conditions such as Parkinson's disease and epilepsy. The researchers describe their device in a paper published November 19 in the Optical Society's (OSA) journal Optics Letters.

The technique of manipulating neurons with light is only a few years old, but the authors estimate that thousands of scientists are already using this technology, called optogenetics, to study the brain. In optogenetics, researchers first sensitize select cells in the brain to a particular color of light. Then, by illuminating precise areas of the brain, they are able to selectively activate or deactivate the individual neurons that have been sensitized.

Ed Boyden, a synthetic biologist at MIT and co-lead researcher on the paper, is a pioneer of this emerging field, which he says offers the ability to probe connections in the brain.

"You can see neural activity in the brain that is associated with specific behaviors," Boyden says, "but is it important? Or is it a passive copy of important activity located elsewhere in the brain? There's no way to know for sure if you just watch." Optogenetics allows scientists to play a more active role in probing the brain's connections, to fire up one type of cell or deactivate another and then observe the effect on a behavior, such as quieting a seizure.

Unlike the previous, 1-D versions of this light-emitting device, the new tool delivers light to the brain in three dimensions, opening the potential to explore entire circuits within the brain. So far, the 3-D version has been tested in mice, although Boyden and colleagues have used earlier optogenetic technologies with non-human primates as well.

Targeting neurons with light

One of the advantages of optogenetics is that this technology allows scientists to focus on one particular type of neuron without affecting other types of neurons in the same area of cortex. Probes that deliver electricity to the brain can manipulate neurons, but they cannot target individual kinds of cell, Boyden says. Drugs can turn neurons on or off as well, he continues, but not on such a quick time scale or with such a high degree of control. In contrast, the new 3-D array is precise enough to activate a single kind of neuron, at a precise location, with a single beam of light.

In an earlier incarnation, Boyden's device looked like a needle-thin probe with light-emitting ports along its length; this setup allowed scientists to manipulate neurons along a single line. The new tool contains up to a hundred of these probes in a square grid, which makes the device look like a series of fine-toothed combs laid next to each other with their teeth pointing in the same direction.

Each probe is just 150 microns across -- a little thicker than a human hair, and thin enough so that the device can be implanted at any depth in the cortex without damaging it. The brain lacks pain receptors, so the implants do not cause any discomfort to the brain itself. As in the earlier model, several light-emitting ports are located along the length of each probe. Scientists can illuminate and change the color of each light port independently from the others.

Adding a third dimension to the probe's light-delivery capabilities has allowed researchers to make any pattern of light they want within the volume of a cubic centimeter of brain tissue, using a few hundred independently controllable illumination points.

"It's turning out to be a very powerful and convenient tool," says MIT professor of electrical engineering Clifton Fonstad, co-lead author of the paper.

Blue for on, yellow for off

Neurons in the brain are not naturally responsive to light, so scientists sensitize these cells with molecules called opsins, light-detecting proteins naturally found in algae and bacteria. Genes for an opsin are transferred to the neurons in a mouse's brain using gene therapy, a process in which DNA is ferried into a cell via a carrier such as a harmless virus. The carrier can be instructed to deliver the DNA package only to certain types of cells.

Different colors of light turn different flavors of opsin on -- blue might cause one opsin to activate a cell, while yellow might cause another opsin to silence it. Neurons that are sensitized with opsins gain these abilities to respond to light.

The response of an individual neuron -- whether to turn on or turn off -- depends on the type of opsin it was sensitized with, and the color of light used to illuminate it. In this way, the tool gives neuroscientists an unprecedented level of control over individual neurons in the brain.

Teams from around the world are currently using the technology developed by Boyden's group to study some of the most profound questions neuroscience tries to answer, such as how memory works, the connections between memory and emotion, and the difference between being awake and being asleep.

"I'm really excited about how the brain computes -- the ebb and flow of consciousness," Boyden says. "We know so little about the brain."

A better understanding of the brain may lead to another benefit of this technology: therapy. If a particular type of cell malfunctions in a particular disease, scientists may be able to use a modified 3-D array as a neural prosthesis that could help to treat neurological conditions. Using light to stop overactive cells from firing might alleviate the uncontrollable muscle action of Parkinson's disease. Cells that cause seizures in the brain could be quieted optically without the side effects of anti-seizure medications. Implants that correct hearing deficiencies are also being explored with this technology.

Although the new device is effective in bringing light to the brain, other challenges remain before optogenetics can be used for medical therapy, Boyden says. Scientists do not yet know for certain whether the body will detect the opsin proteins as foreign molecules and reject them. Gene therapy will also have to prove itself if neurons are to be sensitized with opsin effectively.

"It's a long road," Boyden admits.

Meanwhile, he continues, the demand for the tool is currently higher than his team can supply. Boyden says his group is excited about the possibility of commercializing the new 3-D array, as one potential route that would make the devices available as quickly as possible to the neuroscience community.

Metformin Reduces Hepatic Expression of SIRT3, the Mitochondrial Deacetylase Controlling Energy Metabolism

Metformin inhibits ATP production in mitochondria and this may be involved in the anti-hyperglycemic effects of the drug. Sirtuin 3 (SIRT3) is a mitochondrial protein deacetylase that regulates the function of the electron transport chain and maintains basal ATP yield. We hypothesized that metformin treatment could diminish mitochondrial ATP production through downregulation of SIRT3 expression. Glucagon and cAMP induced SIRT3 mRNA in mouse primary hepatocytes. Metformin prevented SIRT3 induction by glucagon. Moreover, metformin downregulated constitutive expression of SIRT3 in primary hepatocytes and in the liver in vivo. Estrogen related receptor alpha (ERR?) mediates regulation of Sirt3 gene by peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1?). ERR? mRNA expression was regulated in a similar manner as SIRT3 mRNA by glucagon, cAMP and metformin. However, a higher metformin concentration was required for downregulation of ERR? than SIRT3. ERR? siRNA attenuated PGC-1? mediated induction of SIRT3, but did not affect constitutive expression. Overexpression of the constitutively active form of AMP-activated protein kinase (AMPK) induced SIRT3 mRNA, indicating that the SIRT3 downregulation by metformin is not mediated by AMPK. Metformin reduced the hepatocyte ATP level. This effect was partially counteracted by SIRT3 overexpression. Furthermore, metformin decreased mitochondrial SIRT3 protein levels and this was associated with enhanced acetylation of several mitochondrial proteins. However, metformin increased mitochondrial mass in hepatocytes. Altogether, our results indicate that metformin attenuates mitochondrial expression of SIRT3 and suggest that this mechanism is involved in regulation of energy metabolism by metformin in the liver and may contribute to the therapeutic action of metformin.
